Genetic Variability

Abstract: RAPD markers were used to measure the genetic diversity of 119 individuals of Ixodes ricinus collected from Lithuania and Norway. The samples were analysed within and also between the populations. We analysed 74 loci in each of 6 populations. Our results show high levels of diversity within the populations. The percentage of polymorphic loci of the six analysed populations: Birzai, Vilnius, Kretinga, Tjore, Kjosvik and Odderoya were 68.9%, 58.1%, 78.38%, 62.2%, 44.6 % and 68.9%, respectively. The percentage of polymorphic loci in the Lithuanian populations was 93.2%, and in the Norwegian populations 81.08%. The genetic distance ranged from 0.019 to 0.079 within Norwegian populations and from 0.005 to 0.0967 within Lithuanian populations and between the countries from 0.022 to 0.146. Forty-one type and reference strains of Lactobacillus were evaluated using their randomly amplified polymorphic DNA band profiles. Developed bands for each strain were distinct and enabled discrimination. The best correlations were obtained applying the Pearson product moment correlation coefficient (r) together with the unweighted pair group method using arithmetic averages algorithm. All of the strains were clearly differentiated at and below the 72% similarity value. Species discrimination might be possible making use of the distinctly polymorphic bands amplified specific to a strain.

Ten accessions of sunflower were evaluated for achene yield and its related traits. The experiment was laid out following a randomized complete block design with three replications. Observations were recorded on quantitative traits i.e. plant height, leaf area, head diameter, 100 achene weight, achene weight per plant, stem girth, internodal distance and descriptive traits (Leaf shape, head angle, achene colour, achene stripes and seed sizes). The data were subjected to analysis of variance, correlation and path coefficient analysis to evaluate the genetic variability in the breeding material and to estimate association among various traits. The effects of water deficit on photochemical parameters and activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), as well as, cellular damages were investigated in two clones of Coffea canephora differing in drought tolerance. After 6 days without irrigation, predawn leaf water potential fell to −3.0 MPa that was accompanied by the suppression of net photosynthesis in both clones. The variable to maximum chlorophyll fluorescence ratio remained unchanged regardless of the imposed treatments. Both clones showed a similar decline (about 25%) in the photochemical quenching coefficient, but only the drought-sensitive clone exhibited an enhancement (31%) of thermal deactivation under water deficit conditions. The quantum yield of electron transport decreased similarly in both genotypes. Under drought conditions, activities of SOD, CAT and APX increased to a greater extent in the drought-tolerant clone than in the drought-sensitive one. This seemed to be matched with higher protection against oxidative stress, as judged from the lower levels of lipid peroxidation and electrolyte leakage in the drought-tolerant clone. Thus, the ability to increase the antioxidant system activity in order to limit cellular damages might be an important attribute linked to the drought tolerance in C. canephora.

This study uses PCR-derived marker systems to investigate the extent and distribution of genetic variability of 53 Garnacha accessions coming from Italy, France and Spain. The samples studied include 28 Italian accessions (named Tocai rosso in Vicenza area; Alicante in Sicily and Elba island; Gamay perugino in Perugia province; Cannonau in Sardinia), 19 Spanish accessions of different types (named Garnacha tinta, Garnacha blanca, Garnacha peluda, Garnacha roja, Garnacha erguida, Garnacha roya) and 6 French accessions (named Grenache and Grenache noir). In order to verify the varietal identity of the samples, analyses based on 14 simple sequence repeat (SSR) loci were performed. The presence of an additional allele at ISV3 locus (151 bp) was found in four Tocai rosso accessions and in a Sardinian Cannonau clone, that are, incidentally, chimeras.
