To evaluate the EasyCD4 assay, a less expensive method to enumerate CD4+ lymphocytes, in resource-limited settings. Cross-sectional study conducted in the United States and Uganda. We compared CD4+ cell counts obtained on replicate samples from HIV-infected patients by the EasyCD4 assay, a microcapillary flow-based system, and by standard flow cytometry or FACSCount with linear regression and the Bland-Altman method. Two hundred eighteen samples were analyzed (77 in the United States and 141 in Uganda). In the United States, mean +/- SD CD4 was 697 +/- 438 cells/microL by standard flow cytometry and 688 +/- 451 cells/microL by EasyCD4. In Uganda, the mean +/- SD CD4 was 335 +/- 331 cells/microL by FACSCount and 340 +/- 327 cells/microL by EasyCD4. The 2 methods were highly correlated (US cohort, r2 = 0.97, slope = 1.0, intercept = -18; Ugandan cohort, r2 = 0.92; slope = 0.95; intercept = 23). The mean differences in CD4 cell counts were 9.0 and -4.6 cells/microL for the US and Ugandan cohorts, respectively, and they were not significant in either cohort. In the Ugandan cohort, sensitivity and specificity of the EasyCD4 for CD4 below 200 cells/microL were 90% and 98%, respectively. Positive predictive value was 96%; negative predictive value was 93%. Our results suggest that EasyCD4 may be used with high positive and negative predictive value in resource-limited settings.

The issue of improving the method of assessing the effectiveness of future development marketing strategies of cotton ginneries has been studied in the article. The country is taking comprehensive measures to develop the cotton industry, modernize and re-equip ginneries, increase the profitability of production and processing of raw cotton, as well as the competitiveness of its products. At the same time, the real state of af airs in this area indicates the rapid renewal and modernization of production processes, the introduction of advanced technologies and modern methods of corporate governance, the existence of systemic problems and shortcomings that hinder the rapid development of the industry.

The article is devoted to the investigation of methods of countering the spread of extremism and terrorism in the Internet in the European Union and the Russian Federation. The study was carried out on the basis of an analysis of the bulletins issued by the official sources of Europol and in the UK on the issue. The author concludes that it is necessary to apply the administrative and legal mechanisms under consideration to counter extremism and terrorism in the Russian Federation.

The pressure-area relation of coronary arteries provides important information about the mechanical properties of these vessels. In human subjects methodological limitations have precluded measurement of instantaneous compliance and coronary stress in vivo. The purpose of this study was to assess a new method for measuring instantaneous values of coronary artery compliance and wall stress utilizing simultaneously acquired pressure and intravascular ultrasound measurements of vessel area. Ten subjects with coronary artery disease had intravascular ultrasound studies of the proximal left anterior descending or circumflex coronary arteries. A continuous spectrophotometric assay has been developed for detecting beta-glucuronidase activity. In the assay, Para-nitrophenyl beta-D-glucuronide is cleaved to yield a chromophoric product. With the commercial E. coli enzyme, it is demonstrated that the reactions can be continuously monitored by the increase of absorbance at 405 nm. The method is highly sensitive and able to detect less than 1.4 x 10(-4) U/mL of the enzyme activity in solution. Such a new assay offers significant advantages over the existing discontinuous methods and should be useful for both routine enzyme assay and accurate kinetic studies.
