Zusammenfassung
Es werden verschiedene Elutionssysteme für die Trennung und Isolierung der underivatisierten und pyridylethylierten HMW-Untereinheiten von Glutenin durch RP-HPLC an Nucleosil C8 vorgestellt. Während sich für mikropräparative Zwecke ein System auf der Basis von 2-Propanol/Trifluoressigsäure/Acetonitril bewährt hat, erwies sich für die quantitative Bestimmung einzelner HMW-Untereinheiten ein harnstoffhaltiges System mit einem sehr flachen Gradienten wegen seiner hohen Auflösung als besonders brauchbar. 24 Weizensorten mit unterschiedlichen technologischen Eigenschaften wurden quantitativ auf HMW-Untereinheiten analysiert. Die erhaltenen Werte wurden mit physikalischen Daten der Weizensorten (SDS-Sedimentationsvolumen und Dehnwiderstand) korreliert. Es zeigte sich, daß Typ und Menge der HMW-Untereinheiten von Einfluß auf die genannten Größen sind.
Summary
Several elution systems for the separation and the isolation of underivatized and pyridylethylated highmolecular-weight (HMW) subunits of glutenin by RP-HPLC on Nucleosil C8 are presented. A 2-propanol/trifluoroacetic acid/acetonitrile system proved successful for micropreparative purposes, while an urea-containing system with an extremely flat gradient delivered the best quantitative results, because of its high resolving power. The HMW subunits of 24 wheat varieties with different functional properties were quantitatively analyzed. The values obtained were correlated with physical parameters, namely SDS-sedimentation volume and maximum resistance. The results clearly demonstrated the significance of the type, as well as the amount, of HMW sub-units relative to the functional properties.
