    Bu çalışmanın amacı, Fen ve Teknoloji öğretmenlerinin kullandıkları öğretim yöntem ve teknikleri incelemektir. Araştırma Adana ili Seyhan ilçesinde bulunan 10 farklı ilköğretim okulunda görev yapan 11 Fen ve Teknoloji öğretmeni ile gerçekleştirilmiştir. Araştırmada veri toplama aracı olarak "yarı yapılandırılmış görüşme formu" kullanılmıştır. Form yapılandırılmış 6 soru ile yapılandırılmamış 7 soru olmak üzere toplam 13 sorudan oluşmaktadır. Verilerin analizinde nitel araştırma yöntemlerinde kullanılan içerik analizinden yararlanılmıştır. Araştırma sonucunda elde edilen bulgulara göre Fen ve Teknoloji öğretmenlerinin önemli bir kısmı düz anlatım yöntemini mutlaka her konuda kullanmakta ve bu yöntemi soru-cevap, beyin fırtınası, kavram haritası gibi çağdaş öğrenme teknikleri ile desteklemektedir.The purpose of this study is examined the opinions of the Science and Techonology teachers concerning about learning methods. The study has been implemented 11 Science and Techonology teachers working in 10 different primary schools in Seyhan, Adana. -Semi-structured interrogation form- is used in the study. The form consisits of 6 structured and 7 open ended questions, 13 in all. Percentage and frequency are used for the analysis of the answers to structured questions. -Content Analysis Method-, which is used for qualitative methods, are applied for the analysis of the answers to open-ended questions. According to the study results, many teachers have given importance to literacy in every lesson and they support this traditional methods with contemporary technics such as answer question, brain storm, concept maping

    In this research, it was aimed to investigate the production of water-soluble chitosan from the enzymatic deacetylation of chitin by Bacillus sp. (B. cereus (BC) and B. thuringiensis (BT)) strains. Characteristic properties involving molecular weight, degree of deacetylation, and antibacterial activity of chitosan samples were identified. The degree of deacetylation of BC and BT chitosan samples obtained at 393 and 213 ppm amounts was reported as 80.54 and 86.07% by IR spectrum. As well as the degree of deacetylation, molecular weights of samples showed a significant effect on antibacterial activity were 48.27 and 48.46 Da, respectively. Among the tested bacteria, the highest inhibitory effect was recorded to P. aeruginosa and S. epidermis for both chitosan samples. On the other hand, no antibacterial effect on S. aureus, VRE, S. epidermidis, Klebsiella sp., and Salmonella spp were observed.     In this study, multiple antibiotic resistance (MAR) index was investigated in 74 Enterobacteriaceae strains isolated from Seyhan Dam Lake. Plasmids of these strains were isolated and each of these strains plasmid profile was determined. MAR index values changes between 0.29-1.0. It was determined that 39 (52.7%) isolates contain plasmid. By examining the relationships between resistant antibiotic number and plasmid profiles, it was found that Escherichia coli, which was resistant to 7 antibiotics, has not any plasmid, on the other hand another strain of Escherichia coli, which was resistant to 5 antibiotics, has 14 plasmid DNA bands.

    Bu arastirmanin amaci aktif ogrenme yaklasiminin ilkogretim altinci sinif ogrencilerinin fen bilgisi dersine yonelik tutumlari uzerine etkisini belirlemektir. Arastirma 2005-2006 ogretim yilinda Adana ili Seyhan ilcesinde bulunan orta sosyoekonomik duzeyde bir resmi ilkogretim okulunun iki ayri subesinde gerceklestirilmistir. Calismada deney grubunda 34, kontrol grubunda 32 olmak uzere toplam 66 ogrenci yer almistir. Calisma 12 hafta surmustur. Deney grubunda aktif ogrenme yaklasimi kontrol grubunda ise ogretmen merkezli geleneksel ogretim kullanilmistir. Arastirma cesitleme (triangulation) yontemine gore duzenlenmistir. Arastirma sonucunda aktif ogrenme yaklasiminin ilkogretim altinci sinif ogrencilerinin fen bilgisi dersine yonelik tutumlari uzerinde olumlu etkisi (P < .05) oldugu belirlenmistir.

    Özet: Bu çalışmada Akdeniz'den izole edilen 158 adet Enterococcus faecalis bakterilerinin 15 farklı antibiyotiğe ve dört farklı ağır metale karşı dirençliliği sırasıyla agar difüzyon ve agar dilüsyon yöntemleriyle araştırılmıştır. Gentamisine (%98.7), siproflaksine (%77.8), imipeneme (%77.2) ve levofloksasine (%72.8) karşı yüksek antibiyotik dirençliliği bulunurken, en az dirençlilik ise vankomisine (%3.2) minosikline (%13.3) ve kinopristin-dalfopristine (%13.3) karşı bulunmuştur. Çoklu antibiyotik dirençlilik (ÇAD) indeksleri 0.2-0.93 arasında değişmektedir. Birçok izolat farklı ağır metal derişimlerine tolerans göstermiştir. Bu sonuçlar Doğu Akdeniz'in önemli miktarda antibiyotik ve ağır metal dirençliliği taşıyan Enterococcus faecalis barındırdığını ve bununda halk sağlığı açısından potansiyel bir risk taşıdığını göstermektedir. Veriler, 2008–2009 egitim-ogretim yilinda arastirmaci tarafindan gecerlik ve guvenirlik testleri yapilarak hazirlanan saglik bilgisi testi kullanilarak toplanmistir. Arastirmaci tarafindan hazirlanan saglik bilgisi testinin KR-20 guvenirlik analizi yapilmis ve guvenirlik katsayisi 0,84 olarak bulunmustur.     Antibiotic resistant bacteria from commercially marketed fish in Adana, Turkey were investigated. A total of 126 antibiotic resistant bacteria were isolated from gill and intestinal contents of five types of retail fish samples. Viable counts of antibiotic resistant bacteria isolated from gill and intestinal content samples showed high frequencies of resistance to tetracycline, ampicillin, streptomycin, while the proportion of chlorampheni- col resistance was rather low. A high incidence of resistance to tetracycline, ampicillin and cephalothin as well as almost an absence of resistance to amikacin and gentamicin was found among selected isolates which represented the resistant bacterial population. These strains were predominantly resistant to 3 and 4 antibacterials. Isolates from gill exhibited resistance to as many as 7 antibiotics, whereas those from intestinal content were resistant to 5 or fewer antibiotics.     In this study, frequency of antibiotic resistance was investigated in A. hydrophila from hospital effluent water in Turkey. A total of 70 A. hydrophila strains were isolated. The resistance of A. hydrophila isolates to 10 different antibiotics was investigated by agar diffusion method. Results showed that a high incidence of resistance to ampicillin (100.0%), tetracycline (100.0%) and nalidixic acid (87.1%), and a low incidence of resistance to amikacin (15.7%). In the other hand, all isolates found sensitive to nitrofurantoin. Multiple antibiotic resistance (MAR) index values ranging from 0.2 to 0.9. This study suggest that multiple antibiotic resistant A. hydrophila strains easilly recovered from hospital effluent waters in Turkey and these sewages may play as an important reservoirs responsible for infectious diseases pathogen A. hydrophila. At the same time, these bacteria may result in a potential ecological effect.

    The present study was carried out to assess the isolation and determination level of antibiotic resistance patterns of Pseudomonas aeruginosa from the hospital effluent water. A total of 72 P. aeruginosa strains was isolated from the hospital sewages. The resistance of P. aeruginosa isolates to 10 different antibiotics was investigated by agar diffusion method. Results revealed a high incidence of resistance to ampicillin (100.0%), nitrofurantoin (100.0%), nalidixic acid (100.0%) and tetracycline (100.0%), and a low incidence of resistance to meropenem (1.4%) and amikacin (5.6%). In the other hand, all of the isolates found sensitive to imipenem. In this study the multiple antibiotic resistance (MAR) index range found very high (from 0.6 to 0.8). Results show that the hospital effluent water has an important amount of antibiotic multi-resistant P. aeruginosa strains and these bacteria may result in a potential risk for environment.

    The aim of the present study was to determine the level of antibiotic resistance patterns of bacterial isolates from Eastern Mediterranean fish in Turkey. The resistance of 93 Enterobacteriaceae isolates from gills to 10 different antibiotics belonging 7 classes (which most using in Turkey), was investigated by agar diffusion method. A total 13 species of bacteria were isolated: the most common strains were Enterobacter cloacae (49.5%) following by Klebsiella oxytoca (15.1%). Fish may contaminate with discharge of urban or hospital sewages. There was a high incidence of resistance to ampicillin (66.7%), and cefazolin (47.3%), and a low incidence of resistance to kanamycin (1.1%) and cefepime (1.1%). Results show that Eastern Mediterranean fish have significant proportion of antibiotic resistant Gram-negative bacteria, and these bacteria constitute a potential risk for fish population and public health.     ABSTRACT A total of 178 food samples (fresh soft cheese, raw milk, ice cream, sausage, chicken meat and minced beef meat) were investigated for Listeria monocytogenes. Of these, 29 (16.3%) were contaminated. 102 strains of L. monocytogenes were isolated and the susceptibility to 13 different antibiotics was determined. Most of the isolates (57.8%) were resistant to amdinocillin; in contrast, all isolates were sensitive to chloramphenicol. Multiple antibiotic resistance (MAR) index values ranged from 0.21 to 0.43. Results show that antibiotic resistant L. monocytogenes strains were easily isolated from selected foods and antibiotic resistance may become a potential food poisoning concern for public health.

    In this study, it was investigated for cellulase activities of Clostridium strains isolated from soil and rumen. Cellulase isolation was performed and SDS-PAGE profiles were determined from Clostridium CF147 strain which showed the highest cellulase activity. It was determined that cellulase consists of two bands with molecular weight of 81000 Da and 66000 Da, respectively. pH range and thermal stability of this enzyme were determined. Optimum pH value of cellulase was found as 7.0. Cellulase was determined the stable at 55C upto 64%.

    This study aimed to identification and determines the pattern of antibiotic resistance bacteria isolated from shrimps. The susceptibility of 97 bacterial isolates to 16 different antibiotics was investigated by agar diffusion method. The most common species isolated from the samples were Stenotrophomonas maltophilia (19.6%), followed by Acinetobacter lwoffi (14.4%) and Proteus vulgaris (9.3%). There was a high incidence of resistance to cefazolin (100%), cefuroxime (97.9%), nitrofurantoin (97.9%) and ampicillin (96.9%), and a low incidence of resistance to cefepime (7.2%) and meropenem (22.7%) was found among selected isolates which represented the resistant bacterial population. Multiple antibiotic resistance indices ranged from 0.2 to 0.81, suggesting exposure to antibiotic contamination. These results suggest that shrimps of Iskenderun Bay have important proportion of antibiotic resistant bacteria and these bacteria can be responsible a potential risk for public health. At the sa...

    In this study it was aimed to determine the microbial diversity and level of antibiotic resistance patterns of Gram-negative bacterial isolates from the hospital sewages. The 219 Gram-negative bacterial isolates to 16 different antibiotics (belonging 10 classes), was investigated by agar diffusion method. A total of 18 species of bacteria were isolated: the most common strains isolated from all samples were Klebsiella oxytoca (27.4%), Klebsiella pneumoniae (20.5%) and Escherichia coli (20.1%). There was a high incidence of resistance to ampicillin (98.6%), streptomycin (95.9%) and erythromycin (90.0%), and a low incidence of resistance to cefepim (13.2%), imipenem (5.0%) and meropenem (3.2%). 35.6% of all bacteria isolated from hospital sewage were resistant to 9 different antibiotics.     Identification and multiple antibiotic resistances of amphicillin, chloramphenicol, streptomycin and tetracycline resistant gram-negative bacteria that isolated microorganisms from Gölbaşı and Azaplı lakes (Adiyaman) were investigated in this study.     In this study, decolorization of Reactive Blue 220, Acid Red 414 and Basic Yellow 28, decolorization kinetics, and biodegradability of intermediates after ozonolysis were studied. Effect of pH on ozonolysis was studied, and pH 3 was the best among pH values of 3, 7 and 11. The decolorization kinetics of these dyes can be described by a first-order reaction model, and the first-order rate constants of RB 220, AR 414 and BY28 were 0.175-0.1303, 0.164-0.096 and 0.2092-0.1154 min-1, respectively. Total organic removal was determined to be 24.74, 17.15 and 23.37% for RB220, AR 414 and BY 28, respectively, after 30-min ozonation of 800 mg L-1 dye. The respirometric biologic oxygen demand of Pseudomonas putida (30-min ozone treatment of 800 mg L-1 of dye) was 490, 460 and 451 for RB220, AR 414 and BY 28, respectively. Our results showed that ozonation is an effective way to remove the color from aqueous solutions of these dyes, and that the by-products are non-toxic to P. putida.

    In this study it investigated that antibiotic and heavy metal resistance of Gram(-) bacteria from Iskenderun Bay fish’s gills and intestinal content. Bacteria isolated from gill showed high resistance to ampicillin (66.7%) and cefazolin (47.3%) whereas, no isolates showed resistance to imipenem and cefrizoxime. The intestinal bacteria showed the most resistance agains to ampicillin (44%) and cefazolin (36.0%) respectively. A proportion of resistance to chloramphenicol (1.3%) and kanamycin (4.0%) was shown among the intestinal bacteria. These results showed that Iskenderun Bay fish have significant proportion of Gram (-) bacteria with antibiotic and heavy metal resistance and this prompting a health risk to public health for fish consumers.

    In recent years, the studies regarding the anti-Helicobacter pylori activity of medicinal plants have increased considerably. Attention has been given to the screening of medicinal plants all over the world as a means to identify low-cost sources of new drugs against H. pylori, a human gastric pathogen with high morbidity rate. The search for new chemical compounds with bactericidal or bacteriostatic effects against H. pylori is a challenge for the world's medical and scientific communities. Garlic (Allium sativum) has been known one of the most used plants in traditional medicine and the wider cited in the literature for its medicinal properties and proven to have antimicrobial effects. This study shows that garlic extracts produce levels of inhibition to Helicobacter pylori similar to those of the commercial materials. Garlic has a wide spectrum of actions, not only is it antibacterial, but it also has beneficial effects on the cardiovascular and immune systems. The research described here represents an important starting point in the fight against and/or prevention of peptic ulcers, as well as other pathologies associated with helicobacter diseases such as gastric cancer. The extracted material can be used by direct application and involves a simple and economical extraction procedure that avoids isolation or purification techniques.

    The aim of the present study was to determine the level of antibiotic resistance patterns and distribution of heavy metal resistance of bacterial isolates from seawater, sediment and shrimps, and to determine if there is a relationship between antibiotic and heavy metal resistance. We undertook studies in 2007 in the industrially polluted Iskenderun Bay, on the south coast of Turkey. The resistance of 236 Gram-negative bacterial isolates (49 from seawater, 90 from sediment and 97 from shrimp) to 16 different antibiotics, and to 5 heavy metals, was investigated by agar diffusion and agar dilution methods, respectively. A total of 31 species of bacteria were isolated: the most common strains isolated from all samples were Escherichia coli (11.4%), Aeromonas hydrophila (9.7%) and Stenotrophomonas maltophilia (9.3%). There was a high incidence of resistance to ampicillin (93.2%), streptomycin (90.2%) and cefazolin (81.3%), and a low incidence of resistance to imipenem (16.5%), meropenem (13.9%) and cefepime (8.0%). Some 56.8% of all bacteria isolated from seawater, sediment and shrimp were resistant to 7 or more antibiotics. Most isolates showed tolerance to different concentrations of heavy metals, and minimal inhibition concentrations ranged from 12.5 microg/ml to > 3200 microg/ml. The bacteria from seawater, sediment and shrimp showed high resistance to cadmium of 69.4%, 88.9%, and 81.1% respectively, and low resistance to manganese of 2%, 6.7% and 11.3% respectively. The seawater and sediment isolates which were metal resistant also showed a high resistance to three antibiotics: streptomycin, ampicillin and trimethoprim-sulphamethoxazole. In contrast, the shrimp isolates which were metal resistant were resistant to four antibiotics: cefazolin, nitrofurantoin, cefuroxime and ampicillin. Our results show that Iskenderun Bay has a significant proportion of antibiotic and heavy metal resistant Gram-negative bacteria, and these bacteria constitute a potential risk for public health.

    We studied the susceptibility patterns to 15 different antibiotics and six heavy metals in Aeromonas spp. and Pseudomonas spp. isolated from Iskenderun Bay, Turkey (northeast Mediterranean Sea). A high percentage of Aeromonas isolates showed resistance to cefazolin (66.6%) and trimethoprim-sulphamethoxazole (66.6%). Amongst the Pseudomonas isolates, there was a high incidence of resistance to nitrofurantoin (86.2%), cefazolin (84.8%) and cefuroxime (71.7%). Most isolates showed tolerance to different concentrations of heavy metals, and minimal inhibition concentrations ranged from 25 to >3,200 microg/ml. The Aeromonas spp. and Pseudomonas spp. showed high resistance to copper of 98.3% and 75.4%, respectively, and low resistance to lead of 1.7% and 7.2%, respectively. Our results show that antibiotic and heavy metal resistant Aeromonas spp. and Pseudomonas spp. were widespread in Iskenderun Bay in 2007 and 2008. The increasing presence of antibiotic and heavy metal resistant Aeromonas spp. and Pseudomonas spp. may become a potential human health hazard.

    In this study it aimed to determine the microbial diversity, level of antibiotic resistance patterns and distribution of heavy metal resistance of bacterial isolates from the Eastern Mediterranean Sea coast. The resistance of 255 gram-negative bacterial isolates to 16 different antibiotics and to 5 heavy metals was investigated. The most common strains isolated from all samples were Citrobacter koseri (9.0%), Escherichia coli (8.2%) and Pantoea agglomerans (8.2%). Our results revealed a high incidence of resistance to ampicillin (74.0%), streptomycin (70.0%) and cefazolin (48.3%). The multiple antibiotic resistance (MAR) index ranged from 0.2 to 0.75. Isolates showed tolerances to different concentrations of heavy metals. Our results show that the Eastern Mediterranean Sea coast has a significant proportion of antibiotic and heavy metal resistant pathogens, or opportunist gram-negative bacteria, and these bacteria may result in a potential public health hazard.
