Small-Molecule Inhibition of Human Immunodeficiency Virus Type 1 Infection by Virus Capsid Destabilization
ABSTRACT
MATERIALS AND METHODS
Viruses and cell lines.
PCR quantification of reverse transcription.
Antiviral assays.
Assays of HIV-1 capsid stability.
Assay of HIV-1 uncoating in target cells.
RESULTS
Specificity of retroviral inhibition by PF74.
PF74 binds specifically to HIV-1 particles.
PF74 inhibits HIV-1 reverse transcription in target cells.
PF74 destabilizes the HIV-1 capsid.
HIV-1 sensitivity to PF74 is modulated by mutations altering the intrinsic stability of the viral capsid.
PF74 antiviral activity is promoted by cyclophilin A binding to the viral capsid.
