It publishes a family of high-quality scholarly chemistry journals, covering a very broad range of disciplines. Its mission is to evaluate, publish, disseminate, and amplify the scientific excellence of chemistry researchers from around the globe in high-quality publications. It supports its members at every stage of their careers as they strive to solve the challenges that impact humankind. #Metallodrugs are effective weapons in the fight against #cancer. One of the oldest entries into this substance class, #cisplatin, continues to be used to this day. Beyond platinum-containing compounds, which were extensively investigated for their antiproliferative properties, many other metals are also explored for their use in cancer therapy. It is, however, not only the metal but also the organic #ligands that determine the effectiveness and specificity of metallodrugs. María Gil-Moles (CISQ, Universidad de La Rioja) and Concepción Gimeno (ISQCH, Universidad de Zaragoza) highlight and summarize the research on #terpyridine complexes of the coinage metals, copper, silver, and gold. Terpyridines are biologically active themselves, for example as DNA intercalators, and their metal complexes show promising pharmacochemistry. The team reviews structures and mechanisms and identifies open questions for the field. Enzyme engineering is a great way to take the power of enzymes one step further. By so-called “directed evolution”, researchers mimic how evolution works in nature, for example mutating chosen amino acid constituents of proteins that are important for their function. Making smart choices such as limiting possible replacements to such that would also occur in nature can help reducing the vast chemical space that might need to be sampled in such investigations. Kai Schülke, Jana Fröse, Alina Klein, Marc Garcia Borràs (Universitat Girona) and Stephan Hammer (Universität Bielefeld) have used this approach to efficiently engineer transferases that can be used to very efficiently alkylate S-adenosyl-L-homocysteine with high enantioselectivity using commercially available iodoalkane reagents. The team used UHPLC-ESI-MS-based high-throughput screening to quickly identify suitable mutants based on the alteration of “hotspot” amino acid positions identified by molecular dynamics simulations. Their approach allowed hit identification in very short times and could also be used to engineer related systems. 🔮 What will the Sustainable Future smell like? The transition from today’s resource-wasting petrochemical economy towards a 100/100 renewable and biodegradable future is inevitable and has in fact already begun. For some fragrance families, highly performant sustainable alternatives are easier to find than for others, and for some they already existed since their very introduction. The historic routes to synthetic vanillin, which currently celebrates its 150th anniversary, for instance all commenced from renewable starting materials: first coniferin, then guaiacol ex beechwood tar, and finally eugenol, abundantly occurring in clove oil.
