Summary

A simplified method for methyl green pyronin staining is described forTheileria parva andT. annulata in whole salivary glands ofRhipicephalus appendiculatus andHyalomma anatolicum subspecies respectively. The stain gives results comparable with Feulgen staining and can be used after the ticks have been in cold storage for 3 days. There is considerable variability in the rate and intensity of infection of these ticks with theilerial parasites and it is concluded that the method permits large samples (60 ticks per person per day) to be examined to overcome this variability when assessing infection quantitatively.

Résumé

Une méthode simplifiée de coloration par la “methyl green pyronine” deTheileria parva etTheileriia annulata dans l'ensemble des glandes salivaires deRhipicephalus appendiculatus etHyalomma anatolícum est décrite. Cette coloration qui donne des résultats comparables avec celle de Feulgen, peut être utilisée chez des tiques conservées au froid pendant 3 jours. Il existe des variations considérables dans le taux et l'intensité de l'infection de ces tiques par des Theileria. Les auteurs concluent que cette méthode par les nombreux examens (60 tiques par personne et par jour) qu'elle permet fait que l'inconvénient de ces variations peut être surmonté lors de l'estimation quantitative de l'infection.

Resumen

Se describe un método sinplificado de la coloración verde metíl pironina paraTheileria parva yT. annulata, en glandulas salivares de subespecies deRhipicephalus appendiculatus yHyalomma anatolicum respectivamente. La coloración produce resultados similares a la coloración de Feulgen y puede utilizarse despues de guardar las garrapatas en frio por períodos de tres dias. Existe una variabilidad considerable en la proporción e intensidad de la infección de estas garrapatas con parásitos teileriales. Se concluye que este método permite examinar muestras grandes (60 garrapatas persona dia) lo cual conlleva menor variabilidad en estimaciones cuantitativas.
